An increasing number of veterinary practices are performing in
office hematology testing. Historically, this involved time con¬
suming, labor intensive manual methods. A number of auto¬
mated and semiautomated hematology instruments are now
available for analysis of veterinary samples, which, when used
properly, offer more efficient, precise testing than can be achieved
through manual methods. This article reviews manual methods
for performing a complete blood count and describes some of the
available automated hematology instruments. Factors that affect
test results and the importance of a quality assurance program
are also discussed.
Congenital Erythrocyte Enzyme Deficiencies 1003

Congenital hemolytic anemias resulting from pyruvate kinase
(PK), phosphofructokinase (PFK), and glucose 6 phosphate dehy
drogenase (G6PD) enzyme deficiencies in domestic animals art?
described. Persistent methemoglobinemia has been reported in
dogs and cats with methemoglobin reductase enzyme deficiency.
Affected animals have cyanotic appearing mucous membranes
but exhibit no or only mild clinical signs attributable to hypox
emia. Erythrocyte enzyme assays are usually carried out after

acquired causes of hemolytic anemia and methemoglobinemia

The Neutrophil: Function, Disorders, and Testing 1013

The ability of the body to keep harmful materials from interior
spaces is closely linked to survival. One of the critical components
of this process is the neutrophil. This article examines how the
neutrophil moves from rapid circulation in the peripheral blood
to a tissue site of need and how these cells dispatch offending
material. Inherited defects of neutrophil function are discussed
and information on function testing is also provided.
Myelopoiesis and Myeloproliferative Disorders 1023

Myelopoiesis describes the derivation of nonlymphoid cells that
include granulocytes, monocytes, erythrocytes, and platelets from
a common hematopoietic stem cell. Current concepts of hemato
poiesis including stimulating and inhibitory growth factors are
discussed. Disturbances of development in myeloid stem cells
results in dysplastic and neoplastic conditions termed myelopro¬
liferative disorders. A recently devised classification scheme for
these disorders in dpgs and cats provides a uniform manner to
categorize myelodysplastic syndrome, acute myeloid leukemia,
chronic myeloid leukemia, and their subtypes using cytomorpho
logic methods. Emphasis in this article is given to recent studies
in which this classification method can be applied to improve
one s understanding of the pathogenesis of canine and feline
myeloproliferative disorders.
Blood Cell Markers 1043

Cytochemistry and immunocytochemistry are important adjunc
tive technologies to the morphologic characterization of blood
cells and hematopoietic neoplasms. Although cytochemistry is
briefly discussed, the emphasis of this article is on the clinical
application of flow cytometry and leukocyte monoclonal antibod¬
ies in veterinary medicine. Classification and significance of im
munophenotyping in canine and feline lymphomas and immuno¬
deficiencies such as feline immunodeficiency virus and feline
leukemia virus are discussed.
Platelets and Coagulation: An Update 1065

The latest developments in the areas of platelets and coagulation
are presented and related to physiologic and pathologic pro¬
cesses. Platelet integrins, particularly auhp3, axe discussed and

perspectives on coagulation pathways, mechanisms of antiphos

Von Willebrand s Disease in the Dog and Cat 1089

Von Willebrand s disease (vWD) is a bleeding disorder that is
recognized commonly in dogs but rarely in cats. This article
presents the pathophysiology and clinical features of vWD, in¬
cluding a discussion of inherited and acquired types of vWD.
Laboratory evaluation and treatment of vWD are reviewed.
Antithrombotic and Fibrinolytic Factors: A Review 1111

Excessive or nonphysiologic thrombogenesis and fibrinolysis ac¬
companies many diseases. Several specific proteins involved in
the physiologic regulation and maintenance of blood in a fluid
state are reviewed in this article. Assays for these proteins or
evidence of their function (antithrombin III, protein C, protein S,
plasminogen/plasmin, tissue plasminogen activator, plasmino
gen activator inhibitor I, fibrinogen/fibrin degradation products,
and thrombin/antithrombin complexes) are described. Principles,
general methodology, and application in veterinary medicine are
discussed. Although most of the investigative work and knowl¬
edge concerning these proteins and assays has been in human
beings, their use and application in veterinary medicine is becom¬
ing more available in research laboratories at referral centers
and some larger commercial veterinary laboratories. Use and
interpretation of these assays will help clinicians and researchers
better understand pathophysiologic processes occurring in vari¬
ous diseases associated with thrombogenesis and excessive fibri
nolysis.
An Update on Chemistry Analyzers 1129

This update of six chemistry analyzers available to the clinician
discusses several points that should be considered prior to the
purchase of equipment. General topics include how to best match
an instrument to clinic needs and the indirect costs associated
with instrument operation. Quality assurance recommendations
are discussed and common terms are defined. Specific instrument
features, principles of operation, performance, and costs are pre¬
sented. The information provided offers potential purchasers an
objective approach to the evaluation of a chemistry analyzer for
the veterinary clinic.

Pathophysiology of Calcium, Phosphorous, and
Magnesium Metabolism in Animals 1155

The goal of this article is to summarize key features of calcium,
phosphorus, and magnesium pathophysiology and highlight
some of the recent scientific accomplishments in these fields. The
area of calcium physiology has been especially active due to
the discovery of a new calcium regulating hormone, parathyroid
hormone related protein, cloning of the parathyroid hormone
receptor and identification of a cell membrane receptor for ion¬
ized calcium. Advances have also been made in understanding
the role of phosphorus and the active form of vitamin D, 1,25
dihydroxyvitamin D in the pathogenesis of secondary hyperpara
thyroidism in patients with chronic renal failure. The role of
magnesium in many pathologic processes, including cardiac dis¬
ease, is gaining a heightened appreciation due to its function in
many metabolic processes and the development of techniques to
measure ionized magnesium concentrations.
Strong Ion Difference Approach to Acid Base
Imbalances with Clinical Applications to Dogs and Cats 1185

Stewart s strong ion difference, first introduced more than 10
years ago, offers an innovative approach for the analysis of non
respiratory acid base disorders. This article addresses the concept
of strong ion difference and discusses its clinical applications. A
brief review of base excess and anion gap is also included. Clini¬
cal cases are provided to demonstrate a step by step method
using strong ion difference to evaluate nonrespiratory acid base
imbalances.
Invasive Cytology of Internal Organs:

Invasive cytology of the thoracic and abdominal cavities can
provide diagnostic information in a timely manner for the prac¬
titioner. The information depends on obtaining a quality sample
followed by thorough cytologic evaluation. As an adjunct to
the historical and clinical information, cytology is valuable in
establishing a diagnosis or a list of differentials and directing
future diagnostics or therapy. The application of cytology of
internal organs opens a new window for the differential diagnosis
of disease.
Molecular Tools for the Diagnosis of Animal Diseases 1223

This article is a review of the recent advantages in the use of
molecular technology for the diagnosis of animal diseases. The

tools of molecular diagnosis such as cloning techniques, hybrid¬
ization techniques, and the polymerase chain reaction (PCR) are
briefly described. This is followed by a review of how these tools
are currently being used in veterinary medicine to diagnose many
microbial, congenital, and neoplastic diseases in animals. A dis¬
cussion of the advantages and disadvantages of the various meth¬
ods is also provided.
