Induction of multiple ovulations (superovulation) may be a criti¬
cal requirement for successful assisted reproductive technology
in horses. Attempts at superovulation in mares have included
gonadotropin releasing hormone, equine chorionic gonadotropin,
follicle stimulating hormone, equine pituitary extract, and inhibin
immunoneutralization. Equine pituitary extract administration
has been the most successful superovulatory approach used, with
mean ovulation rates from 1.7 to 3.8 ovulations per cycle re¬
ported.
Transvaginal Aspiration 13

This article describes in detail the procedures for collection of
equine oocytes using a transvaginal ultrasound probe. Success in
obtaining oocytes from humans, bovines, and horses are pre¬
sented. The effect of repeated follicular aspiration of both cattle
and horses is reviewed.
Maturation and Fertilization of Equine Oocytes 31

This article reviews the factors affecting the maturation of equine
oocytes in vitro. Culture media, hormones in media, time in

culture, and the addition of somatic cells on the ability of equine
oocytes to mature in vitro are discussed. Equine studies on in
vitro fertilization are also reviewed and assisted reproductive
techniques, such as zona cracking and intracytoplasmic sperm
injection, are discussed.
Gamete Intrafallopian Transfer 47

Gamete intrafallopian transfer (GIFT) involves placement of a
donor s oocyte into a surrogate s oviduct. Fertilization and em¬
bryo development occur within the recipient s reproductive tract.
GIFT provides a viable method to obtain pregnancies from sub
fertile mares for which embryo transfer has been nonproductive.
Currently, pregnancy rates after GIFT have been variable, al¬
though high success rates have been reported. Further technique
refinement should allow GIFT to be used in research and com¬
mercial programs.
Early Embryonic Development and Evaluation of Equine
Embryo Viability 61

This article reviews the process of early embryonic development
in horses and five methods of evaluation the viability of equine
embryos. These are (1) morphologic assessment; (2) development
culture; (3) metabolism of fluorescent substrates; (4) fluorescence
of dead cells; and (5) metabolic tests.
Cryopreservation of Equine Embryos 85

Principles and procedures for cryopreservation of equine em¬
bryos are described. Embryos less than 250 (xM in diameter can
be cryopreserved successfully if glycerol is used as the cryopro
tectant. Cooling is takes place in such a way that most of the
water leaves the cells before intracellular ice forms, and glycerol
is removed after thawing without undue osmotic swelling of cells.
Vitrification procedures also show promise for small embryos.
Satisfactory procedures for cryopreserving embryos of more than
250 (xM in diameter are not yet available.
Use of Semen as Biopsy Material for Assessment of Health

Conventional light microscopic evaluation does not fully utilize
potential indicators in seminal ejaculates for diagnosis of disor¬
ders of the reproductive tract. The technique of evaluation of all

cellular components of semen, as described in this article, utiliz¬
ing both light and transmission electron microscopy is a valuable
diagnostic tool. Compare with other common biopsy procedures,
use of semen as biopsy material is noninvasive, more representa¬
tive than excisional biopsy, less expensive, and helps in the longi¬
tudinal evaluation after a therapeutic regimen.
Methods for Induction of Capacitation and the Acrosome
Reaction of Stallion Spermatozoa 111

A brief description of some of the changes that sperm undergo
during capacitation and the acrosome are reviewed. Methods for
evaluating both capacitation and the acrosome reaction in stallion
sperm are discussed. Finally, the results of current methodologies
for capacitating stallion sperm and inducing the acrosome in vitro
are presented.
Analysis of Stallion Semen and its Relation to Fertility 119

Basic in vitro laboratory semen assays are described, including
methodology to perform these assays as well as what type of
information is obtained from each. The relationship between sem¬
inal quality assays and the fertility of a semen sample also is
discussed.
Cryopreservation of Stallion Spermatozoa 131

This article discusses the advantages and disadvantages of using
cryopreserved semen in breeding programs. Included is a discus¬
sion of the basic principles of freezing cells in general, followed
by a description of the techniques used to freeze stallion sperma¬
tozoa. Possible modifications of freezing protocol, which may
improve cryopreservation of semen that does not survive cryo¬
preservation well, also are discussed.
GnRH Therapy for Subfertile Stallions 149

Idiopathic infertility degeneration has been associated with al¬
tered endocrine function in stallions. Therapy with gonadotropin
releasing hormone (GnRH) has been used in an attempt to correct
these endocrine imbalances and thereby improve fertility. This
article describes the physiologic basis of GnRH therapy and em¬
phasizes the importance of endocrine testing of those stallions for
which GnRH therapy is being contemplated.

Use of Transvaginal Ultrasound Guided Puncture for
Elimination of Equine Pregnancies 161

Twinning is a major cause of abortion in mares. Although early
management of twins is quite successful, twins that remain after
40 days are difficult to manage. This article reviews the current
knowledge of the use of transvaginal ultrasound probe for elimi¬
nation of twins. Several techniques are presented, as well as
discussion on the best time during gestation for this technique to
be used.
