RNAi emerged out of the pioneering work of Andrew Fire and Craig
Mello
in
the nematode,
C. elegans.
Attempting to use antisense
RNA
to knock down
gene expression, they found synergistic effects on gene silencing when anti-
sense and sense
RNA
strands where delivered together. While at first RNAi
seemed a peculiarity of nematodes, dsRNA dependent gene silencing has since
become one of the biggest surprises in the past decade of research in eukaryotic
cells. The core machinery that underlies RNAi is conserved in virtually every
experimental eukaryotic system and has been co-opted in most of them to
trigger gene silencing.
With the advent of RNAi in mammals and the refinement of silencing triggers,
we have reached a point at which any gene in the human or mouse genome can
conceivably be targeted using small dsRNA gene silencing triggers
-
synthetic
small interfering RNAs (siRNAs) or expressed short hairpin RNAs (shRNAs).
In the next decade in the
biomedical
sciences, siRNAs and shRNAs win be em¬
ployed to validate disease models in vitro in
сеД
based systems and in vivo in
rodent and primate systems; to validate drug activities through the remove of
suspected targets; to identify new drug candidates in genome-wide, functional
genomic screens; and finally to combat disease directly, using siRNAs or
shRNAs as therapeutic molecules in the clinic. This volume highlights recent
progress in understanding dsRNA-dependent gene silencing phenomena in
eukaryotic systems ranging from yeast to mammals.
